What is the premise of this protest? What started it?

I think that people support truckers but they do not realize or have forgotten that the trans border issue was and is the initial reason for it. Some folks have turned into something it was not intended to be.
And yes, it is all about the border, Truckers are saying their rights to make a living is denied by the Canadian border restriction but in fact, it is the American border restrictions that keep them out of the US.

Well if the majority of the truckers have been vaccinated, then maybe they're supporting those who do not wish to or cannot take one and they're not necessarily "anti vaxxers" as people claim but people, who for whatever reason, have chosen not to get the jab. I admire the fact that they're willing to go the distance to make their views known rightly or wrongly. That's what we're allowed to do - protest peacefully - and we do!!

Economics, toots. $500 million USD. That's the amount of trade that normally passes daily just along the Ambassador Bridge between Windsor and Detroit. The vast bulk of that is through trucking. In total Canada normally does $1 billion in trade per day with the US. There are no business people, travelers or other groups that have that kind of economic impact crossing the border.
What's fucking sad is the groups that Feds were lauding about being "pandemic heroes" for the 1st year are now being cast as scapegoats.1) There's no medical evidence that suggests truckers have infected Canadians after being in the US. 2) There is absolutely ZERO scientific or medical data that suggests you need a 100% vaccination rate for herd immunity.
When some asshole demands that every single man, woman and child in the country MUST be fully vaccinated, what you have is an asshole with a vested interest in Big Pharma.
