Depends on what kind of train. Go-Transit is still operating through the G20, it's regional though. Basically from Oshawa to Hamilton.

Downtown Toronto is big. The areas you won't be able to access are Front Street, from Yonge to Spadina and north to King. Most everything else in Toronto is business as usual. If you are looking for a hotel, you probably won't find an available room anywhere downtown though.
